Research on Efficacy Analysis for Industrial Use of Sea Urchin

Abstract

In this study, anti-inflammatory, whitening activities of Sea urchin ethanol (SUE) extract were evaluated by use of in vitro assays and cell culture model systems. The cytotoxic effect of SUE extract on cell was examined by the MTT assay. Cells treated with the SUE extract showed a cell viability of 100% or more, therefore, it was determined that there was no cytotoxicity. The anti-inflammatory activities of SUE extract were investigated using RAW264.7 cells treated with lipopolysaccharide (LPS). The effect of SUE extract on nitric oxide(NO) inhibitory activity in LPS-stimulated RAW264.7 macrophages was examined. The SUE extract was used at concentrations of 100, 200, and 400 µg/mL with non cytotoxicity. As a result of measuring the NO production inhibitory ability of SUE exteact on LPS-stimulated macrophages, it inhibited NO production by 3.64%, 12.36%, and 32.89% at concentrations of 100, 200, and 400 µg/mL. Whitening effects of SUE extract were evaluated by melanin contents from Alpha-Melanocyte-stimulating hormone (α-MSH) treatmented B16F10 cells. SUE extract inhibited melanin production by 40.80% and 43.58% at concentrations of 100 and 400 µg/mL. Taken together, these results provide the important new insight that SUE possesses various biological activities such as anti-inflammatory and anti-melanogenic.

본 연구는 성게를 활용한 산업의 활성화 방안을 모색하기 위하여 영덕과 고성에서 구제한성게 에탄올 추출물의 항염증, 미백 효과를 측정하였다. 성게 에탄올 추출물이 세포에 미치는 독성 여부를 특정하기 위하여 MTT assay를 활용하여 조사한 결과, RAW264.7 세포와 B16F10 세포 모두에서 세포 독성이 나타나지 않았다. 세포에 독성이 나타나지 않은 성게 에탄올 추출물의 항염증 활성은 LPS (lipopolysaccharide) 처리한 RAW264.7 세포를 사용하여 조사하였다. LPS로 자극 된 RAW264.7 대식세포에서 SUE-Y 추출물과 SUE-G 추출물이 산화질소(NO) 억제 활성에 미치는 영향을 조사하였다. 두 지역에서 채취한 성게 추출물은 세포 독성이 나타나지 않은 100, 200, 400 µg/mL 농도에서 NO 생성을 평균적으로 3.64%, 12.36%, 32.89% 억제하였다. 항염증과 관련된 유전자 발현 양상을 측정한 결과, 항염증 관련 유전자인 iNOS, COX-2 모두 농도 의존적으로 감소시키는 것을 확인하였다. 또한 성게 추출물의 미백 효과는 Alpha-Melanocyte stimulating hormone (α-MSH)이 처리된 B16F10 세포의 멜라닌 함량 억제능을 측정하여 효능을 평가하였다. 성게 추출물은 100, 200, 400 µg/mL 농도에서 멜라닌 생성을 평균적으로 약 10.80, 20.89, 43.58% 억제하였으며, melanin 생성에 관여하는 tyrosinase 효소 역시 8.32%, 33.83%, 46.15% 감소시키는 것을 확인하였다. 종합하면, 성게 에탄올 추출물이 NO 및 멜라닌 생성, tyrosinase 효소 활성 억제에 우수한 효능을 보이며, 따라서 성게 에탄올 추출물은 항염증 및 미백활성에 우수한 효능을 가진 기능성 식품 소재 및 화장품 소재로서의 개발의 가능성을 보여준다.
